چکیده

Mutants of Bacillus subtilis deficient in urocanase or in imidazolonepropionate hydrolase were isolated. These mutants are unable to use L-histidine as source of carbon or nitrogen for growth. The enzymes are thus essential for the metabolism of histidine. The structural genes specifying these enzymes are closely linked to those specifying the other two enzymes essential for histidine degradation, histidase, and formiminoglutamate hydrolase. The response of these enzymes to induction and catabolite repression is speciCed by a region of the chromosome closely linked to one end of the cluster of structural genes. The gene cluster presumably constitutes an operon.

Biogenic Amine Degradation by Bacillus Species Isolated from Traditional Fermented Soybean Food and Detection of Decarboxylase-Related Genes.

Biogenic amines in some food products present considerable toxicological risks as potential human carcinogens when consumed in excess concentrations. In this study, we investigated the degradation of the biogenic amines histamine and tyramine and the presence of genes encoding histidine and tyrosine decarboxylases and amine oxidase in Bacillus species isolated from fermented soybean food. No ex...

Insights into anti-termination regulation of the hut operon in Bacillus subtilis: importance of the dual RNA-binding surfaces of HutP

The anti-termination protein, HutP, regulates the gene expression of the hut (histidine utilization) operon of Bacillus subtilis, by destabilizing the hut terminator RNA located upstream of the coding region encoding l-histidine degradation enzymes. On the basis of biochemical, in vivo and X-ray structural analyses, we now report that HutP uses its dual RNA-binding surfaces to access two XAG-ri...

Role of CodY in regulation of the Bacillus subtilis hut operon.

Bacillus subtilis mutants deficient in amino acid repression of the histidine utilization (hut) operon were isolated by transposon mutagenesis. Genetic characterization of these mutants indicated that they most likely contained transposon insertions within the codVWXY operon. The codY gene is required for nutritional regulation of the dipeptide permease (dpp) operon. An examination of hut expre...

An alternative bacterial expression system using Bacillus pumilus SG2 chitinase promoter

Background: Chitin is an abundant natural polysaccharide found in fungi, algae, and exoskeleton of insects. Several bacterial species are capable of utilizing chitin as their carbon source. These bacteria produce chitinases for degradation of chitin into N-acetyl-D-glucosamine. So far, regulation of the chitinase encoding genes has been studied in different bacterial species. Among Bacillus spe...

Taxonomic characterization and plant colonizing abilities of some bacteria related to Bacillus amyloliquefaciens and Bacillus subtilis.

The phylogenetic relationships of 17 Bacillus strains isolated from plants and soil were determined from partial sequences of genes encoding 16S rRNA, gyraseA (gyrA) and the cheA histidine kinase. Five strains were closely related to Bacillus subtilis subsp. subtilis, three strains were more closely related to B. subtilis subsp. spizizeni and two strains were identified as B. mojavensis. The re...
